投氟不同时间大鼠血清PTH水平的变化 被引量:3

摘要 目的观察投氟不同时间伴随不同钙营养条件下大鼠血清甲状旁腺激素(PTH)水平的变化。方法48只Wistar大鼠,按体重均匀分成4组。设立常食和低钙两大组,给大鼠饮水投氟(每升水含氟100mg)12周,分别在1周、4周、8周和12周采血。取大鼠一侧股骨采用15%乙二胺四乙酸脱钙,经4%多聚甲醛固定,脱水,石蜡包埋,制作病理切片;利用试剂盒检测不同组大鼠血清的钙和PTH含量。结果光镜下观察到常食投氟组大鼠股骨骨干部位新骨形成明显和未成熟编织骨增多;低钙加氟组观股骨骨干部位骨外膜成骨细胞明显增生,功能活跃呈典型氟骨症病变。生化检测显示氟作用时间的延长可降低血清的钙水平;投氟或/和低钙营养处理均不同程度的促进血清PTH的增多,且随着氟作用时间的延长血清PTH水平提高明显。结论低钙营养可协同氟的骨骼毒性作用,加重骨损害,亚慢性氟中毒促进PTH的分泌,而低钙营养协同氟中毒在短期内即可刺激PTH的分泌,且该刺激作用具有时间依赖性。 Objective To observe the level of serum Parathyroid hormone (PTH) in rots of different fluoride-treated periods under the condition of different caicium nutrition. Methods 48 Wistar rats were divided 4 greups,half of them were treated with sodium fluoride (NaF) through drinking water containing 100 mg F -/L for 3 months, and half of them were fed with low-calcium forage. Isolated rat femur were fixed in 4% paraformaldehyde and decalcified with 15% EDTA in 0.1 M PBS, then dehydrated, embedded in paraffin. The level of serum calcium and PTH were detected by kits. Results It was observed the new and immature bone formation increased in fluoride-treated rats,and osteoblast proliferation and active function in outer membrane of femoral shaft in low calcium plus fluoride-treated rats. Extension of fluoride - treated pereid obviously decreased the level of serum calcium. Overdosal fluoride coordinated with/without low calcium nutrition stimulated PTH secretion. The level of serum PTH significantly higher with the extension of fluoride-treated peroid. Conclusion Overdosal fluoride coordinated with low calcium nutrition aggravated bone tissue lesion; subchronic fluorosis promoted the secretion of PTH, overdosal fluoride coordinated with low calcium nutrition obviously stimulated the secretiou of PTH in short term and showed time-dependent response.
