摘要
目的对4种品牌不同型号的高效液相色谱仪-二极管阵列检测器(HPLC-DAD)的灵敏度进行评价。方法配制不同浓度的维生素A醋酸酯、脱水穿心莲内酯和青蒿素溶液,分别采用Hitachi、Waters、Agilent和Shimadzu公司4种不同型号的HPLC-DAD,比较210、250和328nm波长处信噪比为3时的检测量。结果以上4种HPLC-DAD检测,得到维生素A醋酸酯的检测限分别为(390±s6)×10-3、(480±10)×10-3、(660±10)×10-3和(770±12)×10-3ng,脱水穿心莲内酯的检测限分别是(170.0±2.5)×10-3、(260±4)×10-3、(250±4)×10-3和(450±4)×10-3ng,青蒿素的检测限分别是(530±7)×10-3、(1400±14)×10-3、(1830±24)×10-3和(2320±6)×10-3ng。L-2455DAD检测的3种样品的检测限均显著低于其他仪器的检测结果(P<0.01)。结论L-2455DAD的灵敏度较其他3种型号检测器的灵敏度高。
AIM To evaluate the sensitivity of high performance liquid chromatography-diode array detector (HPLC-DAD) of four manufacturers. METHODS When signal/noise = 3, retinyl acetate, dehydroan- drographolide and artemisinin solutions of different concentrations were used to compare the limit of detection (LOD) on each DAD at 210, 250 and 328 nm, respectively. RESULTS LODs of retinyl acetate on DAD of Hitachi, Waters, Agilent, and Shimadzu were (390 ± s 6)×10^-3, (480 ± 10)×10^-3, (660 ± 10)×10^-3, and (770 ± 12) ×10^-3 ng, respectively. LODs of dehydroandrographolide were (170.0 ± 2.5) ×10^-3, (260 ± 4) ×10^-3, (250 ± 4) ×10^-3, and (450 ± 4) ×10^-3 ng, respectively. LODs of artemisinin were (530 ± 7) ×10^-3, (1 400± 14) ×10^-3, (1 830±24)×10^-3, and (2 320±6)×10^-3 ng, respectively. LOD detected by L-2455 DAD was obviously lower than those of other DADs. CONCLUSION The sensitivity of L-2455 DAD is higher than those of the other three DADs of different manufacturers.
