摘要
目的建立诃予提取物中鞣花酸含量的高效液相色谱(HPLC)测定方法。方法色谱条件Shim—pack CLC—ODS色谱柱(150mm×6mm,5μm),柱温30℃,用CH3OH—EtOAc—KH2PO4/H3PO4(0.05mol/L)(体积比30:2:68)作为流动相,检测波长为254nm。结果鞣花酸在0.268~5.36μg范围内与峰面积呈良好的线性关系。平均加样回收率为98.42%,RSD为1.13%(n=5)。结论该方法快速、简便、准确,可用于诃子中鞣花酸的含量测定。
Objective To develop an HPLC method for the determination of ellagic acid in the extract of Terminalia . Methods HPLC separation was carried out in an Shim -pack CLC -ODS columm (150 mm×6 mm,5 μm)with the mobile phase consisted of methanol ethyl acetate, 0.05 mol/L KH2PO4 -H3 PO4 (34: 2: 64) at a flow rate of 1.0 ml/min. The detection wavelength was 254 nm. Results The linear range was 0. 268 - 5.36 μg/ml for ellagic acid. The average recovery was 98.42% , and RSD was 1.13% ( n =5), Conclusion This method is rapid, accurate and reproducible for determination of ellagic acid in the extract of Terminalia.
