摘要
目的通过检测热休克蛋白70表达的变化,为研究间歇运动抗心脏缺血再灌注损伤的作用机制提供依据。方法SD大鼠随机分成3大组:间歇运动训练组,1次间歇运动组和对照组,间歇运动训练组进行高强度间歇运动训练,1次间歇运动组仅进行一次高强度的间歇运动,对照组不运动,在心脏模型制备时对照组随机分成缺血再灌注对照组和假手术组。运动训练结束后,除假手术组外其余大鼠均采用结扎左冠状动脉制备在体大鼠心肌缺血-再灌注损伤模型。缺血30 min、再灌注40 min后,取左心室心肌组织用蛋白免疫印迹实验和免疫组化方法检测心室肌HSP70的表达。结果经间歇运动训练和1次间歇运动处理的大鼠HSP70的表达显著高于缺血再灌注对照组。结论长期间歇运动训练和1次间歇运动都可能提高缺血再灌注心肌中HSP70的表达。
Objective By detecting expression changes of HSP70, the study provided basis for researching on mechanism of interval exercise training against myocardial ischemia reperfusion injure. Methods Thirty-two SD rats were used and randomly divided into four groups: interval exercise training group (IET, n= 8), once interval exercise group (OIE, n= 8) , the control group (C, n= 8) and sham operated group (S, n = 8). But sham operated group, the other rats were sufferd ischemia reperfusion. Myocardial ischemia reperfusion injure was induced by the occlusion of left anterior descending branch of the coronary artery. After thirty minutes ischemia and forty minutes rcperfusion, activity of myocardial enzymes in plasma and antioxidant enzymes in myocardial tissue were observed. The content of malondialdehyde (MDA) in myocardial tissue were observed too. Results the expression of HSP70 in IET and OIE were remarkably more than sham operated group. Conclusion Both chronic and actue interval exercise can effectively increase expression of HSP70 in isehemia-reperfusion myocardium.
