摘要
目的:建立测定中药和保健食品中低聚果糖(包括蔗果三糖、蔗果四糖、蔗果五糖3种成分)的高效液相含量测定方法。方法:色谱柱Agilent zorbaxNH2(4.6min×250mm,5μm),柱温25℃;流动相水-乙腈(32:68),流速1.2mL·min^-1;视差折光检测器,内部温度35℃。结果:蔗果三糖、蔗果四糖、蔗果五糖的线性范围分别为2.26~22.6μg,2.16-21.6μg,2.10-21.0μg;加样回收率(n=5)分别为96.32%,96.98%,95.67%。结论:本方法简便、灵敏、准确,可用于含低聚果糖中药和保健食品中以上3种成分的含量测定和产品质量控制。
Objective.. To develop an RP HPLC-RID method for determination of fructooligosaccharides (including kestose, nystose, Fructofranosylnystose) in chinese medicine and functional foods. Methods: An Agilent zorbax NH2 column (4. 6 min×250mm, 5μm)was applied; a mobile phase composing of acetonitrile and water(68:32) was used, and the flow rate was 1.2 mL · min^-1 ; A RID detector was applied, and the inner temperature was set at 35℃. Results:The results were obtained that quantitative line range of three fructooligosaecharides was 2.26-22. 6μg,2.16-21.6μg,and 2.10-21.0μg respectively, and the average recovery (n=5) was 96.32%,96.98%,and 95.67%, respectively. Conclusion:A specific, sensitive, simple and reproducible RP- HPLC-RID method was established for the determination of the three ingredients in chinese medicine and functional foods that claimed containing fructooligosaccharides.
