食物中毒变形杆菌的生物学特性及分子分型研究 被引量:7

Study of biological characteristics and genotyping of proteus from food poisoning

摘要 目的:了解食物中毒变形杆菌菌株的毒素性、侵袭力和耐药性情况,比较拮抗试验(the Dienes test)分型和脉冲场凝胶电泳(PFGE)分型用于变形杆菌食物中毒的流行病学意义。方法:对马鞍山市2007年的8起食物中毒检出的28株变形杆菌(25株奇异变形杆菌,3株普通变形杆菌),使用鲎试验(试管法)检测内毒素、刚果红平板法检测侵袭力产生情况,VITEK-32全自动微生物鉴定系统检测菌株药物敏感性,使用Dienes试验分型和PFGE分型分析两者在菌株区分能力上的差别,比较两者在同源性和食物中毒流行病学的意义。结果:从8起食物中毒标本中共检出28株变形杆菌,内毒素检测有21株(阳性率75.0%)为阳性;刚果红试验结果表明,有7株(25.0%)为阳性,其余均为阴性;经耐药分析,菌株对阿米卡星、头孢他啶、庆大霉素、亚胺培南、左旋氧氟沙星、哌拉西林/他唑巴坦、妥布霉素均为100%敏感,对呋喃妥因96.4%耐药。Dienes试验将25株奇异变形杆菌分为4个菌型、3株普通变形杆菌分为同一菌型,而PFGE分型则把25株奇异变形杆菌分为22种带型、3株普通变形杆菌分为2种带型。PFGE对变形杆菌的分型能力高于传统的Dienes试验分型。结论:对于变形杆菌引起食物中毒的溯源追踪上,PFGE分型分析优于传统的Dienes试验分型。 Objective:Understanding of toxin, invasion and antibiotic resistance of proteus from food poisoning, compared the epidemiological purpose of typing by the Dienes test and pulsed -field gel electrophoresis (PFGE) for the proteus from food poisoning. Methods:From eight of food poisoning in Maanshan City in 2007, 28 strains of Proteus were isolated (25 P. mirabilis and 3 P. vulgaris). Limulus Test (test tube) was used for detection of endotoxin, Congo red plate for detecting invasion of strains, VITEK - 32 automatic identification system was used for detecting strains of drug sensitivity. Analysis of discriminatory difference typing by the Dienes test and PFGE genotyping, comparison the homology between strains and epidemiological association of food poisoning. Results:From eight of food poisoning in Maanshan City in 2007,28 Proteus were isolated. 21 are positive for endotoxin detection ( positive rate 75.0% ) Congo red test shows 7(25.0% ) were positive. Analysis of the drug resistance, the strains with 100% sensitive are amikaein, ceftazidime, gentamiein, imipenem, levofloxacin, piperacillin/tazobactam and tobramyein, with nitrofurantoin resistance by 96.4%. The Dienes test divided 25 P. mirabilis into 4 types, 3 P. vulgaris into 1 type, and PFGE analysis divided 25 P. mirabilis into 22 types, divided 3 P. vulgaris into 2 types. PFGE has more Discriminatory powers than the Dienes test. Conclusion: As for tracing sources of food poisoning, typing by PFGE is superior to that of the traditional Dienes test.
