人胃腺癌BGC823细胞Midkine高表达前后基因表达谱芯片分析

摘要 目的:用基因芯片技术分析、筛选Midkine高表达前后胃腺癌相关基因谱的差异表达。方法:建立Midkine高表达BGC823胃腺癌细胞系,提取细胞总RNA,逆转录Cy3、Cy5标记cDNA探针,应用晶芯表达谱芯片,分析差异表达的基因。结果:发现Midkine高表达前后BGC823胃腺癌细胞有显著性表达差异的基因共有550个,其中上调基因407个,下调基因143个。对550条差异表达基因进行初步功能分类分析,结果表明这些基因与Midkine促进胃腺癌发病机制存在相关性。结论:Midkine高表达前后BGC823胃腺癌细胞部分功能基因存在显著的表达差异,多数差异表达的基因参与细胞黏附、细胞骨架形成、细胞周期以及细胞代谢等过程。 Objective: To investigate and screen the difference of gene expression profile in gastric adenocarcinoma BGC823 cell before and after midkine gene over - expression by gene chip technique. Methods : The total RNAs were isolated from BGC823 cells before and after midkine gene over - expression. The eDNA retro - transcribed from equal quantity mRNA were labeled with Cy3 and Cy5 fluorescence as probes. Analysis on differential expression genes was carried out. Results : A total of 550 genes were screened out which expression level changed significantly after midkine over - expression. Among these genes, 407 genes were up - regulated and 103 genes were down - regulated. While these genes were classified initially according to function,it was found that they played important role in pathogenesy of gastric adenocarcinoma promoted by midkine. Conclusion: Significant differences are found in the expression of some functional genes in BGC823 cells before and after midkine over - expression. Most of these genes are involved in cell adhesion, cytoskeleton, cell cycle and metabolism.
