摘要
【目的】寻找一种解决纤维蛋白析出影响微柱凝胶法交叉配血和抗体筛选的方法。【方法】用自行设计的CaCl2离心法和EDTA-Na2法对5份交叉配血中有混合外观凝集(假性凝集)的血样本分别进行处理,筛选出最佳处理条件;另选取一份冷沉淀,按1:5和1:10稀释后用CaCl2离心法和EDTA-Na2法分别模拟进行交叉配血和抗体筛选。【结果】CaCl2离心法的最佳条件是稀释比例1:20,孵育5min,3000r/min离心5min;EDTA-Na2法加入的比例是1:20;均对临床新鲜标本处理结果较好,陈旧标本选用EDTA-Na2法处理较好。【结论】用CaCl2离心法对新鲜抗凝血和用EDTA-Na2法对抗凝和非抗凝血均能很好地解决纤维蛋白析出对微柱凝胶实验的影响,值得在临床推广应用。
[ Objective ] To search a method to solve the effect of micro column gel method in cross match by fibrin educes. [ Methods ] To select five blood samples for cross match with mixed field agglutination in order to screen the best disposal methods by CaCl2 centrifugalization method and EDTA-Na2 method which were designed by ourselves,To select another cryoprecipitate which were diluted 1:10 and 1:15 after mimesis do cross match and antibody screening with CaCl2 centrifugalization method and EDTA-Na2 method. [ Results ] The best methods of CaCl2 centrifugalization methods were dilution radio 1 : 20,incubation 5 min, centrifugalization 3 000 rpm, 5 rain and EDTA-Na2 methods dilution radio were 1 :20; CaCl2 centrifugalization methods are well on fresh samples, EDTA-Na2 methods were good for all blood samples. [Conclusions] Fibrin educe have effect on micro column gel method, It can be resolve by CaCl2 centrifugalization methods for fresh blood samples and EDTA-Na2 methods for anticoagulation or non-anticoagulation blood samples, we recommend to use these methods in clinical.
