扶正解毒汤对镍诱导NRK细胞微核形成的抑制作用 被引量:5

摘要 目的:研究扶正解毒汤(FJD)对硫酸镍(NiSO4)诱导大鼠肾细胞系(NRK)微核形成的抑制作用。方法:以NiSO4、FJD含药血清处理NRK细胞,检测微核率的变化及细胞总的清除自由基抗氧化力(TRAC)。结果:NiSO4(400μmol/L)暴露组细胞微核率、TRAC明显下降,低、中、高剂量FJD血清与NiSO4共处理组细胞微核率显著低于NiSO4暴露组(P<0.05,P<0.01),细胞TRAC则显著提升(P<0.05,P<0.01)。结论:NiSO4可诱发细胞微核形成,FJD对镍诱导NRK细胞微核形成有明显抑制作用,其机制可能与抗氧化应激有关。 Objectives: To study the inhibitory effect of Fuzheng Jiedu Decoction(FJD) on the micronuclei formation induced by nickel sulfate(NiSO4) in rat renal cell line(NRK).Methods:Cultured NRK cells were exposed to NiSO4(400μmol/L) and FJD drug serum.Micronuclei(MN) were scored in 1000 NRK cells and cellar total radical-trapping antioxidative capacity(TRAC)was measured.Results: Compared with NiSO4 group,the MN frequencies decreased and TRAC increased in groups treated with both FJD and NiSO4 significantly ( P 〈 0.05, P 〈 0.01 ). Conclusion : FJD could suppress nickel-induced micronuclei formation in NRK ceils which was probably related to its antioxidant property.
