摘要
为了探讨白三叶遗传转化中最佳的卡那霉素筛选体系,将白三叶种子置于不同浓度的卡那霉素溶液中,处理不同时间.结果表明:卡那霉素对白三叶种子的发芽率和发芽势没有显著影响,但对主根的生长和侧根的形成有显著的抑制作用.结合根系生长、叶色、植株长势等指标,确定卡那霉素处理白三叶种子的最佳浓度为50 mg/L,最佳处理时间为6-8d.
In order to find the most effective selecting system of kanamycin in the transgenic process of white clover, the seeds were placed on different kanamycin concentrations with different treatment times. The results indicated that kanamycin had no effect on germination energy and germination percentage, but inhibits top root growth and lateral root development. With other factors taken into consideration such as root growth, leaf color and plant growth vigor, 50mg/L kanamycin concentration and 6-8 days treatment were the most effective selecting system for transgenic white clover.
