摘要
单核细胞增生性李斯特氏菌是一种能够引起人畜共患的食源性致病菌。为了鉴定该菌,研究以单核细胞增生性李斯特氏菌的李氏溶血素基因(Hly)为靶基因,通过环介导等温扩增技术(LAMP)对本基因进行了扩增,并对反应时间、反应温度、镁离子浓度等扩增条件进行优化。结果表明:在Mg2+浓度为4.0mmol/L时,63℃孵育60min,扩增可达最佳效果。
Listeria monocytogenes is a kind of food-borne pathogenic bacteria which can cause human and animal diseases. In order to detect Listeria monocytogenes, we chosed the hemolysin gene (Hly) of L.monocytogenes as target gene, and amplified the gene by Loop-mediated isothermal amplification (LAMP). Meanwhile, the reaction conditions were optimized including temperature, time and Mg^2+ concentration etc. The result indicated that the best amplification conditions was 63 ℃ for 60 min when Mg^2+ concentration at 4.0 mmol/L.
