摘要
目的:建立紫外分光光度衍生化法测鲜牛乳中链霉素残留量的最佳实验条件。方法:将试样与衍生化试剂邻苯二甲醛(OPA)进行胺缩醛反应,即化学衍生处理(CFD法),生成具有特定波长的紫外吸收的衍生产物,测定其吸光度,求出样品中链霉素残留量,并与高效液相色谱法(HPLC)比较。结果:链霉素在0.05-10.0μg/ml的浓度范围内具有良好线性关系(r=0.9983),3种添加浓度0.25、1.50、5.0μg/ml的平均回收率为86.6%(n=5),RSD为2.6%;样品含量测定结果与高效液相色谱法比较,无显著性差异(P〉0.05)。结论:UV-CFD法操作简便、快速,准确度与精密度较高,设备要求低,易推广,可用于鲜牛乳中链霉素残留的检测。
Objective: To establish a method of derivative UV spectrophotometry for the determination in the best condition experiment of streptomycin resduce in fresh milk. Methods : Streptomycin and the derlvatization reagent o - phthalaldehyde ( OPA ) occur Amine Acetalization( CFD method) generating derivative product with UV absorption and determined the absorbance in special wave length, obtained streptomycin residues in samples then compare with high - performance liquid chromatography (HPLC). Results:The streptomycin in concentrations of 0.05 - 10. 0 μg/ml range have a good linear relationship, correlation coefficient r = 0. 9983 ; at 0. 25, 1.50, 5.0 μg/ml, recovery rates of the method was 86. 6% ( n = 5) and relative standard deviation (RSD) was 2. 6% ; the sample assay results compared with the high - performance liquid chromatography, no significant difference ( P 〉 0. 05 ) was found. Conclusion : The method is simple, repid, highly selective and sensitive, of no complex equipment and easy to promote, so it can be used for rapid detection of streptomycin residues in fresh milk.
