摘要
目的确定超声波法提取平菇多糖的最佳工艺。方法采用超声波法提取平菇多糖,利用苯酚硫酸法显色,在490 nm处测定吸光度值,并以正交试验设计优化工艺条件。结果平菇多糖的最佳提取工艺条件为料液比1:30、超声时间60 min、提取温度40℃、超声功率为175 W。结论本实验平菇多糖多糖提取方法及含量测定操作简便,结果准确,重现性好,可用于平菇多糖的提取与含量测定。
Objective To study the optimum extration process of polysaccharides from Pleurotus ostreatus by ultrasonic technology. Methods Polysaccharides was extracted by ultrasonic technology from Pleurotus ostreatus, colored by phenol-sulfuric acid method and determined by spectrophotometry at 490 nm. The optimum technological conditions was determine by the orthogonal tests. Results The optimum technological conditions were deduced : the ratio of water to raw material was 1 : 30, ultrasonic time was 60 min, extraction temperature was 40℃ and ultrasonic power was 175 W. Conclusion The method was proved to be simple, accurate and relia- ble, and could be used to extract and determine the polysaccharides from Pleurotus ostreatus.
