针刺对乳腺增生模型大鼠雌激素、雌激素受体及其信号传导的干预作用 被引量:24

Effect of Acupuncture on Estrogen,Estrogen Receptor and Estrogen Signal Transduction in Rat Model of Hyperplasia of Mammary Glands

摘要 【目的】观察针刺对雌激素介导的乳腺增生模型大鼠的影响。【方法】选用SD雌性未孕大鼠,随机分为4组:正常组、模型组、针刺组(电针膻中、三阴交穴)、西药组(灌胃三苯氧胺0.5 mg.kg-1.d-1);除正常组外,其他组均采用肌注己烯雌酚(剂量为0.5 mg.kg-1.d-1)及黄体酮(剂量为4 mg.kg-1.d-1)方法复制乳腺增生模型;造模成功后针刺组及西药组治疗30 d,采集标本,检测各组大鼠体质量,第2对乳头高度,血清雌激素(E2)水平,雌激素受体(ER)、细胞外信号调节激酶(ERK)、内皮型一氧化氮合酶(eNOS)的表达及乳腺组织形态学变化。【结果】各组大鼠治疗前后体质量、治疗前乳头高度差异无显著性意义(均P>0.05),治疗后模型组乳头高度显著增加(P(0.01),各治疗组可显著降低乳头高度(均P(0.05);模型组出现明显乳腺增生形态学改变,各治疗组可显著减少乳腺小叶腺泡数、腺泡腔直径及导管直径(均P(0.05或P(0.01);模型组大鼠血清E2水平,乳腺组织ER、ERK、eNOS蛋白表达阳性细胞数显著增加(均P(0.01),各治疗组可显著降低上述指标(P(0.05或P(0.01)。【结论】针刺治疗乳腺增生的作用可能与其能调节雌激素信号转导通路,抑制雌激素的促乳腺增生效应有关。 Objective To observe the effect of acupuncture on rat model of estrogen-mediated hyperplasia of mammary glands.Methods SD female non-pregnant rats were randomized into 4 groups: normal group,model group,acupuncture group(electro-acupuncture on acupoints of Tanzhong and Sanyinjiao),and western medicine group(gastric gavage of tribenzylamine 0.5 mg·kg-1·d-1).Except the rats in the normal group,rats in other groups received muscular injection of diethylstilbestrol(0.5 mg·kg-1·d-1) and progesterone(4 mg·kg-1·d-1) to induce hyperplasia of mammary glands.After the modeling,acupuncture group and western medicine group received the corresponding treatment for 30 days.After treatment,rats body weight and the height of the second pair of nipples were examined;serum estrogen(E2) level,and expression levels of estrogen receptor(ER),extracellular regulated kinase(ERK) and epithelial nitric oxide synthase(eNOS) were detected,and the mammary histological features were observed.Results The differences of body weight and nipple height were insignificant between the four groups before treatment(P〉0.05).After treatment,the nipple height was increased in the model group(P〈0.01 compared with the normal group),but was decreased in the two treatment groups(P〈0.05 compared with the model group);hyperplasia of mammary glands occurred in the model group,and acini number in lobules of the mammary gland,acinar cavity diameter and acinar ductal diameter were reduced in the two treatment groups(P〈0.05 or P〈0.01);serum E2 level and the number of mammary cells with ER,ERK and eNOS expression positive were increased in the model group(P〈0.01 compared with the normal group),while were decreased in the two treatment groups(P〈0.05 or P〈0.01).Conclusion The therapeutic mechanism of acupuncture for the treatment of hyperplasia of mammary glands is probably related with the regulation of estrogen signal transduction system,and with the inhibition of estrogen in promoting mammary proliferation.
