复方甘草酸苷对哮喘大鼠的免疫调节作用 被引量:4

摘要 目的探讨复方甘草酸苷对哮喘大鼠的免疫调节作用。方法Wistar大鼠随机分成健康对照组(Ⅰ组)、哮喘模型组(Ⅱ组)、复方甘草酸苷哮喘治疗组(Ⅲ组)、地塞米松哮喘治疗组(Ⅳ组),采用卵清蛋白(ovalbumin,OVA)氢氧化铝混合液腹腔注射致敏和OVA超声雾化吸入激发建立哮喘大鼠模型。所有大鼠在末次激发后6h取中心静脉血及左肺支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF),行细胞计数与分类,并测定细胞因子IL-4和IFN-γ含量。结果Ⅱ组静脉血及BALF中中性粒细胞、淋巴细胞、嗜酸性粒细胞计数和IL-4含量显著高于Ⅰ组、Ⅲ组和Ⅳ组(P<0.01),IFN-γ含量显著低于Ⅰ组、Ⅲ组和Ⅳ组(P<0.01);②Ⅲ组与Ⅰ组相比,静脉血及BALF中中性粒细胞、淋巴细胞、嗜酸性粒细胞计数和IL-4含量升高,IFN-γ含量降低,但均无统计学意义(P>0.05);③Ⅲ组与Ⅳ组相比,静脉血及BALF中中性粒细胞、淋巴细胞、嗜酸性粒细胞计数和IL-4含量升高(P>0.05),IFN-γ含量显著升高(P<0.01)。结论复方甘草酸苷能明显促进Th1细胞因子IFN-γ的产生,减少Th2细胞因子IL-4的产生,减轻哮喘大鼠气道炎症反应,其机制可能与调节Th1/Th2细胞免疫平衡有关。 Objective To evaluate the effects of compound glycyrrhizin on immune regulation in asthmatic rats.Methods Forty Wistar rats were randomly divided into health group(group Ⅰ),asthma group(group Ⅱ),compound glycyrrhizin treating group(group Ⅲ)and dexamethasone treating group(group Ⅳ)(n=10 in each group).Group Ⅱ,group Ⅲ and group Ⅳ received intraperitoneal injection of ovalbumin(OVA)and aluminum hydroxide gel for sensitization and OVA aerosol by ultrasonic nebulization for challenge.In addition,group Ⅲ received intraperioneal injection of 200 μg of compound glycyrrhizin 1 hour before OVA aerosol challenging every time, and group Ⅳ were injected with 100 μg of dexamethasone contrast with compound glycyrrhizin. Group Ⅰ were injected and nebulizated with normal saline. All rats were sacrificed 6 hours after the last challenging. Venous blood and bronchoalveolar lavage fluid (BALF) were collected, the cells were counted and detached,and the levels of IL-4 and IFN-γ were determined. Results OThe count of neutrophilic granulocytes, lymphocytes, eosinophils and the level of IL-4 in group Ⅱ were significantly higher, and the level of IFN-γ in group Ⅱ was significantly lower than those in group I, group Ⅲ and group Ⅳ (P〈0. 01). (2)The level of IL-4 and the count of neutrophilic granulocytes, lymphocytes, eosinophils in group Ⅲ were higher than those in group Ⅰ, and the level of IFN-γ was lower than that in group Ⅰ, but there was no significant difference (P〉0. 05). (3)The level of IL-4 and the count of neutrophilic granulocytes, lymphocytes, eosinophils in group Ⅲ were higher but not significantly different (P〉0. 05) than those in group Ⅳ, but the level of IFN-γ was significantly higher than group Ⅳ (P〈0. 01). Conclusion Compound glycyrrhizin increased the secretion of IFN-γ (Th1 cytokine), and decreased the secretion of IL-4 (Th2 cytokine) significantly, which alleviated the airway inflammation in asthmatic rats. The mechanism may be related to the balance regulation of Th1/Th2 cell-mediated immunity.
