摘要
目的观察游离脂肪酸(FFA)、IL-6对人成熟脂肪细胞中NYGGF4基因表达水平的调节作用。方法体外培养人前体脂肪细胞,采用1-甲基-3-异丁基黄嘌呤(MIX)、胰岛素、地塞米松、罗格列酮方案诱导人前体脂肪细胞分化为成熟脂肪细胞,以1mmol/L混合FFA(油酸、亚油酸、月桂酸、肉豆蔻酸和花生四烯酸)、30μg/L人重组IL-6干预成熟脂肪细胞6、24h,均设未干预细胞为对照,采用实时荧光定量RT-PCR技术检测干预后成熟脂肪细胞中的NYGGF4基因mRNA表达水平。结果1mmol/L混合FFA干预人成熟脂肪细胞6h,NYGGF4基因mRNA相对表达量为0.00151±0.00024,与对照组(0.00114±0.00028)比较无显著性差异,干预时间延长至24h,NYGGF4基因mRNA相对表达量为0.00191±0.00019,显著高于对照组(0.00142±0.00019)(P<0.01);30μg/LIL-6干预人成熟脂肪细胞6hNYGGF4基因mRNA相对表达量为0.00058±0.00031,显著低于对照组(0.00114±0.00028)(P<0.05),干预24h,NYGGF4基因mRNA相对表达量为0.00123±0.00019,与对照组(0.00142±0.00019)比较无显著性差异。结论FFA可显著上调人成熟脂肪细胞中NYGGF4基因的表达,IL-6对人成熟脂肪细胞中NYGGF4基因表达具有一过性下调作用。
Objective To explore the regulative role of free fatty acid(FFA) and interleukin- 6 (IL- 6) on NYGGF4 gene expression in matured human adipocytes. Methods Human preadipocytes were cultured in vitro and differentiated into the matured adipocytes with Insulin,3- isobutyl -methyl xanthine (MIX), Dexamethasone and Rosiglitazone. Fully differentiated adipocytes (Day 18 ) were treated with 1 mmol/L FFA cocktail composed of oleic ,linoleic ,lanric, myristic and arachidonic, meanwhile with 30 μg/L IL - 6 for 6 hours and 24 hours and untreated adipocytes as control group. Total RNA from these adipoeytes were extracted and the levels of NYGGF4 mRNA expression were evaluated by real - time quantitative RT - PCR. Results Treatment of the human matured adipocytes with 1 mmol/L FFA for 6 hours increased NYGGF4 gene mRNA expression. However,this effect showed no statistical significance[ (0. 001 51 ± 0. 000 24) vs (0. 001 14 ±0. 000 28 ) 1. The upregulating effect of FFA on the NYGGF4 gene mRNA expression tended to be enhanced with the elongation of time course. The level of NYGGF4 gene mRNA expression in human matured adipocytes treated with 1 mmol/L FFA for 24 hours was much higher than untreated adipocytes [ (0. 001 91 ±0. 000 19) vs (0. 001 42 ±0. 000 19 ) P 〈 0.01 ] ;30 μg,/L IL- 6 treatment for 6 hours significantly decreased NYGGF4 gene mRNA expression in human matured adipocytes [ (0. 000 58 ±0. 000 31 ) vs (0. 001 14 ±0. 000 28) P 〈0.05]. The negative effect of IL - 6 was reduced after IL - 6 exposure longer. When stimulated with IL - 6 for 24 hours, there was no significantly difference between the IL- 6 treated and untreated -adipocytes in the level of NYGGF4 gene mRNA expression[ (0. 001 23 ± 0. 000 19) vs (0. 001 42 ±0. 000 19) P 〉 0. 05 ]. Conclusions FFA can upregulate the expression of NYGGF4 gene mRNA in human matured adipocytes,and IL- 6 may have a transient inhibitory effect.
