摘要
报道了以荧光素为指示剂,荧光法间接测定痕量SCN-的新方法.在pH=5.5~7.5的中性缓冲溶液中,I2与荧光素反应,使荧光素的荧光猝灭.当加入SCN-时,SCN-和I2反应使体系荧光增强,该体系激发波长、发射波长分别为494nm和515nm,SCN-含量在1.0~60μg/L范围内呈良好的线性关系,检测下限为1.0μg/L.该方法灵敏度高,选择性好,用于吸烟人和非吸烟人唾液中痕量SCN-测定,结果令人满意.
A highly sensitive and selective fluorimetric new method has been developed for the determination of trace amounts of thiocyanate with fluorescein as the fluorogenic reagens (λ ex =494nm,λ em =515nm) at pH=5.5~7.5. In a neutral medium,I 2 reacts with fluorescein to produce a non fluorescence compound.If thiocynate are present,the fluorescence intensity of the solution increases with the increasing concentration of SCN - when approriate amounts of I 2 and fluorescein are added. The calibration range is linear for 1.0~60μg/L for thiocyanate and the detection limit is 1.0 μg/L, it has been applied to the determination of thiocyanate in the saliva with satisfactory results.
