摘要
目的观察丁酸钠对膀胱癌细胞生长的影响。方法不同浓度的丁酸钠干预后,采用^3H—TdR掺入试验比较了膀胱癌BIU-87和E—J细胞的生长曲线变化,并采用流式细胞术研究了丁酸钠对膀胱癌细胞周期的影响。结果1mmol/L浓度组在各观察时间点均未显示出对E—J细胞的抑制;5、10mmol/L在各观察点显示出明显的生长抑制作用,各组之间差异有统计学意义(P〈0.01);随着丁酸钠浓度的升高,越来越多的BIU-87和E—J细胞细胞被阻滞在G0/G1期。结论丁酸钠对肿瘤细胞增殖的抑制作用具有浓度、时间依赖性,这种作用是通过G0/G1期阻滞实现的。
Objective To investigate the effect of sodium butyrate on growth of bladder cancer cells. Methods Make a comparison among the growth curve changes of BIU-87 cell and E-J cell of bladder cancer after the interference of sodium butyrate with different concentrations by applying 3H-TdR incorporation test, and adopt flow cytometry to conduct a research on the influence of sodium bntyrate on the cell cycle of bladder cancer. Results 1 mmol/L concentration group hash' t shown the restraint on BIU-87 cell and E-J cell in each observation time spots;the 5mmol/L and 10 mmol/L concentration groups have shown obvious growth restraint and the difference is significant (P 〈 0.01 ) ;along with the rising sodium butyrate concentration, more and more BIU-87 cells and E-J cells arc blocked in G0/G1 phrase. Conclusion The sodium butyrate restrain the growth of bladder cancer cells with concentration and time dependence, which is achieved through the cell cycle blockade in G0/G1 phrase.
