大黄酸对输尿管梗阻大鼠肾组织纤维化的保护机制 被引量:34

Protection Mechanisms of Rhein on Rat Kidney Fibrosis with Unilateral Ureteral Obstruction

摘要 目的:观察大黄酸(RH)对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的作用及机制。方法:将雄性SD大鼠随机分成三大组:假手术对照组(Sham组,n=6),UUO模型组(结扎左侧输尿管制作UUO模型,n=18),UUO大黄酸干预组(UUO+RH组,n=18)。除Sham组外,其余2组根据不同的观察时间细分为3,7,14d亚组(n=6)分别测定大鼠左肾皮质匀浆中脂质过氧化物标志物MDA以及抗氧化酶CAT和SOD含量。免疫组织化学用SP法检测肾脏TGF-β,bFGF的表达。结果:大黄酸干预组与UUO模型组相比有显著性差异。大黄酸能减少单侧输尿管梗阻侧肾皮质脂质过氧化物的产生,同时增加抗氧化酶的含量,减少TGF-β1和bFGF表达。结论:大黄酸通过阻止TGF-β信号传导,从而阻断肾间质的纤维化,发挥肾保护作用。 Objective: To observe the effects and its mechanism of rhein on renal interstitial fibrosis with unilateral ureteral obstruction (UUO) in rat. Method: Male SD rats were randomly divided into three groups: sham operation group (Sham group, n = 6),UUO model group( ligation of the left ureter UUO model production, n = 18 ), UUO rhein intervention group (UUO + RH group, n = 18). In addition to Sham group, the other 2 groups according to different observation time 3,7,14 d divided into subgroups (n = 6), the rat renal cortex homogenate lipid oxide markers MDA and antioxidant enzymes CAT and SOD content were measured. The kidney TGF-β, bFGF expression were measured by Immunohistoehemistry SP method. Result: The rhein intervention group compared with the UUO model group were significantly different. The rhein can reduced the generation of lipid peroxide of renal cortex in side of unilateral ureteral obstruction, while the content of antioxidant enzymes was increased, TGF-β1 and bFGF expression was reduced. Conclusion: The rhein can prevent TGF-β signal transduction, thereby blocking tubulointerstitial fibrosis.
