摘要
目的:考察啤酒花茎叶不同提取部位的体外抗肿瘤活性及其物质基础。方法:使用乙醇对啤酒花茎叶进行提取,然后分别用石油醚、乙酸乙酯和正丁醇依次对乙醇提取部位进行分段萃取,采用MTT法测定各提取部位对体外培养人肺腺癌细胞系A549和肝癌细胞系HepG2的抑制作用,并应用各种色谱技术从乙酸乙酯提取部位分离化学成分。结果:啤酒花茎叶的乙醇提取部位对A549和HepG2细胞生长具有明显的剂量依赖性抑制作用,乙酸乙酯提取部位对两种细胞的半数抑制浓度(IC50)小于其他提取部位;从乙酸乙酯提取部位分离得到5个化合物,并经1H-NMR波谱分析及与相应标准品的薄层色谱比较,确证其结构分别为β-谷甾醇、β-胡萝卜苷、槲皮素、槲皮苷和金丝桃苷。结论:啤酒花茎叶的乙酸乙提取部位是其主要的抗肿瘤活性部位,这可能与从该部位分离到的5个化合物相关。
Objective: To investigate in vitro anticancer activities of different extraction parts from the stems and leaves of Humulus Lupulus L. and the chemical constituents therein contributable to anticancer activity. Methods: The extraction from the stems and leaves of Humulus Lupulus L. with ethanol and then from the ethanol extraction part in turn with petroleum ether, ethyl acetate and n-butanol was conducted. The inhibitory effects of these extraction parts on A549 and HepG2 cells were measured by MTT assay.The chemical constituents in the ethyl acetate extraction part were isolated by chromatographys. Results: The ethanol extraction part showed significant dose-dependent inhibition of growth of the two tumor cell lines, and the ICs0 of the ethyl acetate extraction part against both cell lines was markedly lower than those of other extraction parts. 5 compounds,β-sitosterol,β-daucosterol, quercetin, quercitrin and hyperin, were isolated from the ethyl acetate extraction part and their structures were verified by 1H-NMR and TLC ( compared with the corresponding reference substances). Conclusion: The ethyl acetate extraction part from the stems and leaves of Humulus Lupulus L. is the main anticancer active part, which could be associated with the isolated 5 compounds therein.
