摘要
以HCV蛋白疫苗PCX3为抗原,分别加入Al(OH)3佐剂,CpG ODN佐剂,以及两种佐剂联合作用的复合佐剂,对雄性六周龄BALB/c小鼠进行常规免疫.与其他免疫组相比,Al(OH)3和CpG ODN复合佐剂与PCX3抗原联合作用组产生的特异性IgG(1×106)和IgM(1×105)抗体滴度最高、持续时间最长;自然杀伤效应(CTL)实验显示该组小鼠诱发的CTL杀伤效应(40.91%)显著高于其他免疫组(P<0.05);流式细胞仪对小鼠淋巴细胞亚群进行分型分析发现:从第2周开始,该组小鼠体内CD4+/CD8+细胞比值(3.26)显著高于PCX3抗原单独作用组(P<0.05).结果表明,Al(OH)3和CpG ODN复合佐剂可以从体液免疫和细胞免疫两方面对HCV蛋白疫苗PCX3的免疫效果进行增强,并且具有协同效应.
The antigen-HCV protein vaccine PCX3 with Al(OH)3 adjuvant,CpG ODN adjuvant,and the combination of them were separately used to immunize six-week-old male BALB/c mice.Compared with other groups,the group which has PCX3 antigen and the combined-adjuvant of Al(OH)3 and CpG ODN,was detected the highest specific IgG(1×106) and IgM(1×105) antibody titer and the longest lasting time by ELISA method;The CTL response(40.91%) significantly higher than other groups(P〈0.05);Since the 2th week,the ratio of CD4+/CD8+ T lymphocyte was significantly higher than the group with PCX3 antigen alone with cyto metry technology(P〈0.05).The results demonstrated that the combined-adjuvant of Al(OH)3 and CpG ODN can enhance the immune effect of HCV protein vaccine PCX3 in both humoral immunity and cellular immunity aspect,and has a synergistic effect.
