Expression of profibrotic growth factors and collagen in neurofibroma tissue

摘要 目的:观察结缔组织生长因子(CTGF)在神经纤维瘤组织中的表达情况及与其上、下游效应基因表达的关系,探讨CTGF在神经纤维瘤组织中的促纤维化机制。方法:收集神经纤维瘤组织、瘢痕疙瘩组织及正常皮肤组织标本,免疫组化检测CTGF的表达,逆转录-聚合酶链式反应(RT-PCR)方法检测组织CTGF、TGF-β1、Ⅰ型胶原(ColⅠ)及Ⅲ型胶原(ColⅢ)mRNA表达,Western blotting检测组织CTGF蛋白的表达。结果:免疫组织化学染色发现,CTGF在瘤细胞中有强阳性表达,在汗腺和皮脂腺细胞中有阳性表达。神经纤维瘤组织及瘢痕疙瘩组织中CTGF、TGF-β1、ColⅠ和ColⅢ的mRNA过表达,而正常皮肤组织无或仅有极弱表达。Western blotting结果显示神经纤维瘤组织有明显的CTGF蛋白表达条带。结论:在神经纤维瘤发病过程中,CTGF可能是重要的调控因子之一,与TGF-β1协同促进细胞外基质合成。 Objective To detect the expression of connective tissue growth factor(CTGF) and its upstorm and downstorm effectors in neurofibroma tissue,and to investigate the profibrotic effect of CTGF in neurofibroma tissue.Methods Specimens of neurofibroma tissue,keloid and normal skin were collected.The distribution and expression of CTGF was measured by immunohistochemistry.The mRNA expression of CTGF,TGF-β1,ColⅠ and ColⅢ was detected by reverse transcription-polymerase chain reaction(RT-PCR).Expression of CTGF protein was measured by western blotting.Results The CTGF was most significantly expressed in tumor cells,and also expressed in sweat gland and sebaceousgland cells.The mRNA of CTGF,TGF-β1,ColⅠ and ColⅢ in neurofibroma tissue were over-expressed,whereas that in normal skin tissue was none or rare.Western blotting shows that CTGF protein was over-expressed in neurofibroma tissue.Conclusion CTGF may by one of the main regulating factors during the profibrotic process of neurofibroma.CTGF may promote the production of extracellular matrix in coordination with TGF-β1.
