摘要
目的本实验采用酶组织化学法观察前列地尔预处理对大鼠心肌缺血再灌注损伤是否有保护性,并探讨其可能机理。方法选用275±25g的SD雄性大白鼠,随机分为6组,每组8只。假手术对照组(A),心肌缺血再灌注模型组(B),经典缺血预处理组(C),前列地尔脂肪乳剂早期保护组(D),前列地尔脂肪乳剂延迟保护组(E),前列地尔脂肪乳剂在缺血前72h应用组(F)。采用酶组织化学方法观察心肌过氧化氢酶及琥珀酸脱氢酶表达。结果与模型组相比,经典缺血预处理组、前列地尔预处理早期保护组及延迟保护组心肌过氧化氢酶及琥珀酸脱氢酶明显升高(p<0.05),且早期保护作用强于延迟作用。结论前列地尔预处理对大鼠心肌损伤具有早期和延迟保护作用。
Objective To investigate the protective effects of isehemic preconditioning with alprostadil E1 on myocardial ischemie-reperfusion models of rats. Methods Rats were randomly divided into six groups: sham operated group, ischemic-repeffusion model, isehemie preconditioning group, prostaglandin E1 early and delayed pretreatment protection groups, 72h alprostadil E1 pretreatment group. The content of eatalase (CAT) and sueeinate dehydrogenase(SDH) was measured in myoeardium by enzymohistoehemical technique. Results The eontenls of CAT and SDH in isehemie precondition, prostaglandin E1 pretreatment groups were significantly higher than those in sham operated group and isehemie group, and higher in prostaglandin E1 earlier protective group than in delayed protective group. Conclusion Prostaglandin E1 pretreatment has earlier and delayed protective effects on isehemie reperfusion injury.
