绿茶提取物对模拟日光照射所致光损伤的防护作用 被引量:2

摘要 目的:观察绿茶提取物对模拟日光照射所致光损伤的防护作用。方法:招募20名健康志愿者,在其背部选取7个非曝光部位。阴性对照部位不予照光亦不涂抹检测物质,阳性对照部位予1.5倍最小红斑量的模拟日光照射但不涂抹检测物质,另有5个部位分别涂抹基质及2%~5%绿茶提取物并于半小时后予照光。照射前后测量皮肤红斑指数(erythma index,EI)。于照射后取皮肤病理进行免疫组化染色,观察表皮厚度(epidermal thickness,ET)、基质金属蛋白酶(metalloproteinases,MMP)-2、-9含量。结果:在阴性对照部位EI差值、ET、MMP-2评分、MMP-9评分分别为8.2、60.4μm、1.8和2.1;在阳性对照部位上述指标分别显著升高至168.0、105.4μm、2.4和2.6;而涂抹2%或3%绿茶提取物的照光部位上述指标升高程度显著低于其它照光部位,分别为131.5~143.4、79.5~83.1μm、1.8~1.9和2.1(P<0.05)。结论:2%~3%绿茶提取物可有效地防护模拟日光照射引起的光损伤。 Objective To investigate the protective effects of green tea extracts on the skin irradiated by solar simulator ultraviolet irradiation (ssUVR). Methods Twenty healthy volunteers were enrolled in this study. Seven sites on the nonexposed back skin were marked for the experiment. Negative control site received neither ssUVR nor test materials, and positive control site received ssUVR but with no test materials. Another 5 sites were applied with base cream or 2% -5% green tea extracts followed by ssUVR at a dosage of 1.5 minimal erythema dose. Pre- and after-irradiation, erythema index (El) was measured. Skin biopsies were taken after the last ssUVR, and immunohistochemistry was used to detect the epidermal thickness (ET) and the expression of metalloproteinases (MMP)-2, -9. Results On negative control site, the El difference value, ET, MMP-2 score and MMP-9 score were 8.2, 60.41~m, 1.8 and 2.1, respectively. On positive control, these index obviously increased to 168.0, 105.4tJm, 2.4 and 2.6, seperately. Whereas on 2%N3% green tea extracts-treated sites, those index increased least comparing to other irradiated sites (P〈0.05), with the values of 131.5-143.4, 79.5N83.1pm, 1.8-1.9 and 2.1. Conclusions Green tea extracts in the concentration of 2%-3% were essential for protection against ssUVR-induced photodamage.
