镉、铅、汞和氟对驱蚊草增殖与生根过程中POD和EST同工酶表达的影响

摘要 以驱蚊草为试材,利用组织培养技术,采用聚丙酰胺凝胶电泳方法,研究了镉、铅、汞和氟对驱蚊草POD和EST同工酶谱带的影响。结果表明,在生根和增值过程中,POD同工酶带活性均呈现出低-高-低的变化趋势,其中生根过程中不同处理浓度之间以镉和汞对POD同工酶带活性变化影响最大,铅最小;而增殖过程中以铅和氟对POD同工酶带活性变化影响最大,汞最小,并且铅和氟处理可增加POD同工酶表达种类。在生根过程中,随着处理浓度的增加,镉和汞对EST同工酶活性均有所下降,而铅和氟对EST同工酶活性均有所增强;在增殖处理中铅对EST同工酶带活性变化最大,汞最小。一定浓度的镉、铅、汞和氟胁迫处理,均增加EST同工酶的表达种类。 Using tussue culture , the effect of the cadmium, the lead, the mercury and the fluorine on POD and EST isozymes MozzieBuster was studied by PAGE. The results shwed that POD isozymes belt activity presents tendency of he low-high-low changes in rooting and multiplication ,in rooting processes activity of POD isozymes changes biggest with the cadmium and the mercury of different concentrations, on the contrary which of lead is smallest. Whereas ,in multiplications processes activity of POD isozymes changes biggest with the lead and the fluorine of different concentrations, which of the mercury is smallest and the lead and fluorine treatment can increase the POD isozymes expression types. In the certain scope, the EST isozymes activity declines along with increasing of the cadmium and the mercury concentrations, but treatment of the lead and the fluorine EST isozymes activity enhances in rooting process. Change of EST isozymes belt activity is biggest in lead of different concentrations, and which of the mercury is smallest in the multiplication process. On stress of the cadmium, the lead, the mercury and the fluorine in the certain concentration the expression types of EST isozymes were all increased.
