摘要
目的制备格拉司琼纳米乳(granisetron nanoemulsion,GRN-NE)并考察其理化特征。方法采用伪三元相图法筛选处方,以十四酸异丙酯(Isopropyl myristate,IPM)为油相、卵磷脂(Phosphatidyl choline,PC)和吐温80(Tween80,T80)为表面活性剂、乙醇(Alcohol,Alc)为助表面活性剂,按GRN:IPM:PC:T80:Alc:water=0.05:1.15:1.4:1.25:14.6的配比,采用自微乳化法制备GRN-NE。用pH计、黏度计、电导仪、折光仪、激光粒度测定仪、TEM和DSC对其进行表征,考察稳定性。结果制备的GRN-NE pH为6.83,黏度为21.70 mPa·s,电导率为0.42 ms·cm^(-1),折光率为1.3771 n_D^(25),透射电镜下纳米乳乳滴呈球形,外观圆整,分布均匀,平均粒径为8.64 nm,Zeta电位为-28.3 mv,在常温和低温条件下稳定性良好。结论本方法工艺简单可行,制备的纳米乳质量稳定,具有一定的开发价值。
Objective To prepare granisetron nanoemulsion( GRN -NE), and investigate its physicochemical characteristics. Methods The formulation and matching of prescription were screened with pseudo-ternary phase diagram. The GRN-NE was prepared with GRN-isopropyl myristate-phosphatidyl choline-tween80-alcohol-water and .their ratio of 0.05: 1.15:1 : 4: 1.25: 14. 6 through self-microemulsification method. The digital pH meter,viscometar,conductimeter,rcfractometer,particle size radiomcte, transmission electron microscope and differential scanning calorimetry were utilized to investigate the characterization of the GRN-NE. And also the stability of GRN-NE was studied. Results The GRN-NE with pH value of 6.83, viscosity of 21.70 mPa · S, conductivity of 0.42 ms · cm^- 1, refractive index of 1. 3771 nD 25, average diameter of 8.64 nm and zata electric potential of - 28.3 mv were obtained. The appearance of GRN-NE was globular and well-distributed under TEM, it was stable under the common and low temperature conditions. Conclusion These resuhs indicated that the obtained GRN-NE with simple and feasible preparative method and stable physico - chemical property had potential development value.
