摘要 目的:观察leptin对人成熟脂肪细胞中NYGGF4基因表达的调节作用。方法:体外培养人前体脂肪细胞,在诱导人前体脂肪细胞分化成熟的基础上,应用100ng/ml人重组leptin分别干预成熟脂肪细胞6、24h,采用实时荧光定量RT-PCR技术检测成熟脂肪细胞中NYGGF4基因mRNA的表达水平。结果:100ng/ml leptin干预人成熟脂肪细胞6h后,NYGGF4基因mRNA的相对表达量为0.000476±0.000178,显著低于对照组0.00114±0.000275,P<0.05;干预24h NYGGF4基因mRNA的相对表达量为0.000835±0.000211,也显著低于对照组0.001419±0.000193,P<0.05。结论:leptin能显著下调成熟脂肪细胞中NYGGF4基因的表达。 Objective:To explore the regulative role of leptin on NYGGF4 gene expression in matured human adipocytes. Methods: Human preadipocytes were cultured in vitro and differentiated into matured adipocytes. The matured adipocytes were treated with 100 ng/ml leptin for 6 hours and 24 hours, respectively. Total RNA from these adipocytes was extracted and the levels of N3fGGF4 mRNA expression were evaluated by real-time quantitative RT-PCR. Results:Treatment with 100 ng/ml leptin for 6 hours or 24 hours significantly decreased NYGGF4 gene mRNA expression in human matured adipocytes (P 〈 0.05). Condusion:Leptin could downregulate the expression of NYGGF4 gene mRNA in human matured adipocytes.
