唐菖蒲花粉离体萌发的液体培养基研究 被引量:11

摘要 以唐菖蒲栽培品种"粉友谊"的新鲜花粉为试验材料,采用花粉离体培养法,研究蔗糖、H3BO3、CaCl2所组成的基本培养基配方不同组分浓度对唐菖蒲花粉萌发率的影响,并在此基本培养基配方的基础上,研究Mg2+、Zn2+、Fe3+、Cu2+和K+离子对花粉萌发的影响。蔗糖、H3BO3、CaCl2单因子和正交试验结果表明,蔗糖和H3BO3对唐菖蒲花粉萌发有显著影响,蔗糖是花粉萌发的必需物质。低浓度的Mg2+、Fe3+和K+对花粉的萌发有促进作用,离子浓度过高对花粉萌发产生抑制作用。而Cu2+、Zn2+在所设定的浓度范围内均表现出抑制作用。唐菖蒲花粉培养液最适配方:蔗糖100 g.L-1+H3BO3100 mg.L-1+CaCl2150 mg.L-1+Fe2(SO4)350 mg.L-1+KNO350 mg.L-1+MgSO4.7H2O 100 mg.L-1;最适培养温度为25℃,且光照、黑暗的环境条件对花粉萌发无显著影响。 The fresh pollens of Gladiolus hybrid "Friendship" were employed as experimental material to study the effects of different concentrations of sucrose,boron,calcium and five kinds of salt ions on vitality test of gladiolus pollen.The results of single factorial experiment and the orthogonal design showed that the concentration of sucrose and boron in culture medium had significant effects on pollen germination.The application of lower concentration of Mg2+,Fe3+ and K+ induced the germination of gladiolus pollen, while the treatment of Cu^2+ and Zn^2 + inhib-ited the the germination of gladiolus pollen regardless of the concentration of their application. The optimum culture solution was sucrose 100 g·L^-1 + H3BO3100 mg·L^-1 + CAC12150 mg·L^-1 + Fe2(SO4)350 mg·L^-1 + KNO350 mg·L^-1 + MgSO4·7H2O 100 mg·L^-1. The optimum temperature was 25 ℃. The effects of light or dark treatment on the germination of gladiolus pollen were not significant.
