摘要
建立了病猪肺脏组织的处理方法和DNA提取方法,根据猪肺炎支原体的P36基因设计一对引物,建立猪肺炎支原体PCR检测方法,并对该方法进行了特异性和敏感性试验,使用建立的方法检测了临床样品。结果表明,建立的检测方法能成功从猪支原体肺炎可疑肺组织中扩增出目的条带,成功建立了猪支原体肺炎可疑肺组织PCR检测方法,为该病的诊断提供了条件。
The treatment methods of lung organization and the suspicious culture of Mycoplasma hyopneumoniae (Mhp) were established. According to the P36 gene of Mhp, two primers were designed. And the specificity and sensibility assay were performed. The clinical samples were detected. The result showed that the gene could be specifically amplified from Mhp and be different from the genes of Mycoplasma hyorhinitis, Mycoplasma flocculate and Mycoplasma gallisepticum. In addition, the method could detect at least 4.26 ng of Mhp genome. The PCR method for detecting Mhp was developed with high specificity and sensibility.
