摘要
目的探讨外源AMP对IEC-6细胞增殖和凋亡的影响。方法IEC-6细胞分成4组,1个对照组和3个实验组(A1、A2、A3组),实验组分别添加10、100、1000μmol/L的AMP,在第2、3、4、5、6天分别检测细胞增殖情况。收集培养48 h后的细胞,检测细胞周期,细胞凋亡和细胞内Ca2+水平。结果不同浓度的AMP都能够抑制IEC-6的增殖(P<0.01),改变细胞周期中各期细胞的相对构成,使得S期细胞数量减少,G0/G1期细胞数量增多。AMP能够诱导细胞凋亡,尤其是1 000μmol/L的AMP,使得细胞凋亡率明显高于对照(P<0.01)。添加AMP后Ca2+水平与对照组略有提高,但差异无统计学意义(P>0.05)。结论外源AMP能抑制IEC-6细胞增殖,影响细胞周期,促进细胞凋亡,且作用与其补充浓度有关,但与钙离子浓度无关。
Objective To explore the effect of adenosine monophosphate (AMP) on the proliferation and apoptosis of IEC-6 cells. Methods IEC-6 cells were divided into 4 groups, one control group and three experimental groups. Experimental groups were treated with 10,100,1 000 μmol/L of AMP, respectively ( as A1 ,A2,A3 groups). The proliferation of cells was determined on 2 -6 d. The apoptosis, cell cycle, and Ca^2+ content were determined after 48 h culture. Results The supplementation of AMP reduced cell proliferation significantly ( P 〈 0.01 ), while the distribution of cell cycle altered with AMP treatment as well. The percentage of cells in S-phase reduced and replaced by a corresponding increase in G0/G1 phases. The treatment of AMP, especially at 1 000μmol/L, induced apoptosis. There was no significant difference in Ca^2+ content ( P 〉 0.05 ) before and after AMP treatment. Conclusion AMP can inhibit proliferation of IEC-6 cells, influence cell cycle and promote apoptosis, and the effect of AMP depends on its concentration, but has no relation with Ca^2+ content.
