摘要
目的:比较糖尿病肾病大鼠和中药复方肾消康治疗组大鼠肾脏组织中基因表达的差异。方法:实验大鼠分为正常组、模型组、治疗组。从肾脏组织中抽提mRNA,由纯化的总RNA合成双链cDNA,并将其纯化,以生物素标记cRNA合成,纯化和质控体外转录(IVT)产物,片断化cRNA,然后进行杂交、洗脱、染色、芯片扫描并用软件进行统计分析,筛选出差异表达数据。不同肾脏组织样本用灰度扫描图和杂交信号强度散点图表示。结果:仅对筛选出差异表达基因谱中的Cxcl12进行报告。糖尿病肾病模型组肾脏组织基因Cxcl12表达上调,中药复方肾消康治疗组肾脏组织基因Cxcl12表达下调。结论:Cxcl12在糖尿病状态下肾脏也有表达,并且表达上调,而中药复方肾消康可使其下调,这也是中药复方对糖尿病性肾组织损伤具有保护作用的机制之一。
Objective: To study the difference of renal tissues gene - expression between diabetic nephropathy rats and rats therapied by herb prescription. Methods: Experimental rots were made of: normal.model and therapy. To abstract mRNA into from renal tissues firstly ; to make purification total RNA into suplex cDNA; to sublimate cDNA ; to compose cRNA which labeled with biotin;to sublimate and control transcript product in vitro;to fragment cRNA ;to hybridize and wash and stain and scan finally, cDNA probes were hybridized with gene chip and make a result through scan and software analysis. To select those data. Different renal tissues samples were labeled by two kinds of grey degree scan and scatter graph. Results:Get different expression Gene -Cxcll2. The expression of diabetic nephropathy gene -Cxcll2 is up,while the expression of rats renal gene - Cxc112 therapied by Shenxiaokang is down. Conclusion:The paper only report one gene - Cxcll2 of rats renal gene expression profiles, renal tissues gene - Cxcll2 is also expressed when suffering from diabetic and the number of expression is more than normal. While Shenxiaokang can make the number of this gene expression less significantly. This is one of protection mechanisms of diabetic renal tissues damage therapied by herb prescription.
