摘要
目的:观察苏木、苏木+黄芪对荷瘤小鼠脾树突细胞的干预作用。方法:复制C57BL/6小鼠Lewis肺癌动物模型,动态观察苏木、苏木+黄芪对抑瘤率、肺转移抑制率、荷瘤小鼠体重、生存期的影响,接种后第15天分离纯化脾树突细胞,流式细胞仪检测树突细胞表型。结果:荷瘤晚期各中药组小鼠体重均高于对照组,但各组比较无统计学差异;各实验组各时间点小鼠瘤重均低于对照组,其中苏木组第6天、黄芪+苏木组第10天瘤重与荷瘤对照组相比存在差异(P<0.05),各实验组抑瘤率随荷瘤天数的增加总体呈降低趋势;各中药组小鼠20天肺转移抑制率均优于对照组,其中黄芪+苏木组肺转移抑制率为77.75%;中位生存期分别为黄芪+苏木组(39天)>苏木组(35天)>荷瘤对照组(24天);各组小鼠脾DCCD11c、CD80、CD86百分比由低至高为荷瘤对照组<苏木组<黄芪+苏木组<正常对照组,正常对照组(51.64±3.08、64.65±4.05、73.00±2.92)各指标均明显高于荷瘤对照组(27.11±4.24、29.14±5.92、5.41±2.11)(P<0.01)。黄芪+苏木组CD80(56.45±20.20)与荷瘤对照组相比存在差异(P<0.05),CD11c(43.69±6.34)与荷瘤对照组相比差异显著(P<0.01),荷瘤对照组、苏木组、黄芪+苏木组CD86表达无差异。结论:荷瘤小鼠脾DCCD11c、CD80、CD86表达均明显低于正常对照组,中药干预能上调DC表面标志表达,益气活血组方苏木+黄芪作用优于单纯活血药苏木。
Objective ： To observe the effect of dendritic cells in spleen of Lewis lung cancer - bearing mice intervened by Sumu and Sumu ＋ Huangqi. Methods ： Lewis lung cancer model in mice was established and treated with Sumu （Sm） and Huangqi ＋ Sumu （Hq ＋ Sm）. The tumor inhibiting rate,the nhibiting rate of lung metastasis, change of weight and survival time was observed and the phcnotype of dendritic ceils in spleen was quantified by flow cytometry （ FCM ） （15d）. Results： The body weight was increased in every treatment groups in late stage, while there was no difference a- mong all groups. The decrease of tumor weight was obvious in Sm group（6d） and Hq ＋ Sin（10d） （ P 〈 0.05 ）. The inhib- iting rate of lung metastasis was increased in two treatment groups especially the Hq ＋ Sin group （P 〈 0.05 ）. The median survival times were 39d in Hq ＋ Sm group ,38d in Hq group ,35d in Sm group and 24d in control group. The proportion of CD11c ＋ , CD80 ＋, CD86＋ cell population of dendritic cells in negative control group were obviously higher than those in positive control group. The proportion of CD80 ＋ cell population in Hq ＋ Sm group was higher than that of positive control group. The increase of the proportion of CD11 c ＊ ceil was obvious in Hq ＋ Sm group. There was no difference of the proportion of CD86 ＋ eel/population among negative control group, Sm group and Hq ＋ Sm group. Corwlusion： The Qi - defteiency and blood stasis compound formula （ Hq ＋ Sm） can up - regulate the phenotype of dendritic ceils in spleen of Lew is lung cancer- bearing mice and this effect was better than that of its component blood- stasis medicine（Sm）.
