Cx26在桥本甲状腺炎甲状腺上皮细胞中的表达

摘要 目的探讨Cx26在桥本甲状腺炎甲状腺上皮细胞中的表达,探索其在自身免疫性甲状腺疾病的发病机制、转归及预后中所起的作用。方法采用二步法免疫组织化学技术(S-P法),测定甲状腺腺瘤旁正常甲状腺组织(对照)16例和桥本甲状腺炎(HT)30例甲状腺滤泡上皮细胞中Cx26的分布及表达情况。结果①对照组和HT组甲状腺滤泡上皮细胞均有Cx26的表达,两者既定位于甲状腺滤泡上皮细胞的细胞质中,又定位于细胞膜上;②对照组和HT组甲状腺上皮细胞Cx26表达阳性率,分别为68.75%(11/16)、20.00%(6/30)。表达强度明显低于对照组(Z值为3.123,P<0.001)。结论Cx26可在人甲状腺滤泡上皮细胞上表达;Cx26在AITD的表达强度存在异质性,这种缝隙连接蛋白表达的异质性可能与AITD的发生、发展及预后有关。 Objective To investigate the expression of Cx26 in Hashimoto disease and the role of the connexin in the de- velopment and turnover of autoimmune thyroid diseases. Methods S - P immunohistochemical method was used to de- termine the expression and the distribution of Cx43 in 16 cases with normal thyroid tissues of thyroid adenoma and 30 cases with Hashimoto, s thyroditis (HT).Results (1) Cx26 express in the thyroid epithelial cells in control and HT group, and the connexin protein is localized in the cytoplasm and the plasma membrane at the points of neighboring cell contact. (2) The positive rates of Cx26 in thyroid epithelial cells from control and HT group were 68.75% (11/16), 20.00% (6/30), and the expression levels of Cx26 in HT was significantly lower than those of normal thyroid tissue(Z, 3. 123, P 〈 0.001 ). Conclusion There might be Cx26 expression in the human thyroid epithelial cells, and there are heterogenicity in levels of Cx26 expression in autoimmune thyroid diseases. The different expressions may play an important role in the development and turnover of autoimmune thyroid diseases.
