摘要
目的:研究西归中总氨基酸含量的测定方法。方法:采用单因素试验对提取条件进行选择,采用分光光度法测定西归中总氨基酸含量,茚三酮显色后检测波长为570nm。结果:总氨基酸的最佳提取条件为物料比1∶15,乙醇浓度为70%,水浴回流提取2次,每次2h;标准曲线方程A=0.031 3C+0.038 9,r=0.999 7,酪氨酸含量在2~12μg/ml范围内线性关系良好。平均回收率为99.44%,RSD为2.47%(n=9)。结论:此方法简便,准确性好,为西归药材的质量控制提供了一个定量方法和参考依据。
Objective: To study the method of determination of total amino acid in seselopsis. Methods: Adopting one factor trial to choose the extract condition; after detection, using Absorption Spetrometry to determine the content of total amino acids at 570.00 nm with ninhydrin as developer. Results: The optimum condition to extract total amino acids is Seselopsis: alcohol=1:15,70% for alcohol; two bath reflux extractions for two hours; the standard curve equation of A=0.0313C+0.0389,r=0.9997,and a good linear correlation was found in range of 2 to 12ug/ml within the tyrosine concentration. The average recovery was 99.44%, ( RSD =2.47% ,n =9 ). Conclusion: The method is simple, accurate, which provides quantitative method and reference for quality control of seselopsis..
