摘要
目的通过观察支气管哮喘小鼠模型T淋巴细胞亚群调节性T细胞(Treg)、Th17的变化,探讨淋巴细胞亚群Treg和Th17在哮喘发病中的作用。方法采用卵清蛋白致敏方法建立支气管哮喘小鼠模型,Balb/c小鼠16只随机分为正常对照组和哮喘组各8只。采用酶联免疫吸附试验(ELISA)技术检测小鼠肺泡灌洗液(BALF)、血清中白细胞介素-10(IL-10)、白细胞介素-17(IL-17)水平;HE染色评价各组小鼠气道炎症程度;采用流式细胞仪检测各组Treg、Th17细胞占CD4+细胞百分率情况。结果哮喘组淋巴细胞亚群CD4+CD25+Foxp3+细胞占CD4+细胞百分率显著低于正常对照组(P<0.01);CD4+IL-17+细胞占CD4+细胞百分率显著高于正常对照组(P<0.01)。CD4+CD25+Foxp3+细胞/CD4+IL-17+细胞比值与嗜酸粒细胞、中性粒细胞、淋巴细胞数呈负相关(r=-0.894,-0.957,-0.886;P均<0.01)。结论Treg/Th17在小鼠哮喘模型中存在失衡表达,提示Treg/Th17失衡参与了哮喘的发病过程,是对哮喘发病免疫机制的重要补充。
[Objecfive] By observing the evolvement of T lymphocyte subgroups Treg and Th 17 in asthmatic mouse models, this study was designed to research the role of Treg and Thl7 in the asthma. [Methods] The Blab/c mice asthma model was induced by ovalbumin (OVA) with classic method. Sixteen female mice were randomly divided into control group and asthmatic group. The level of IL-10, IL-17 in mice bronchoalveolar lavage fluid (BALF) and serum was measured by enzyme-linked immunoserbent assay (ELISA). The airway inflammation was evaluated by HE staining. The percentage of CD4^+CD25Foxp3^+ and CD4^+L-17^+ cells were assessed by intracellular cytokine staining and flow cytometric analysis. [Results] The frequencies of CD4^+CD25Foxp3^+ cells in asthmatic group were significantly lower than those of control group (P 〈0.01). The frequencies of CD4^+IL-17^+ cells in asthmatic group were significantly higher than those of control group (P 〈0.01). The ratio of CD4^+CD25^+Foxp3^+ cells/CD4^+IL-17^+ cells was correlated negatively with the number of eosinophils, lymphocyte and neutrophils. [ Conclusions] Th17/Treg functional imbalance exists in asthma, suggesting a potential role for Th17/Treg imbalance in the onset and development of asthma.
