摘要
目的:研究髓细胞触发受体2(TREM 2)在脂多糖(LPS)致急性肺损伤(AL I)小鼠肺泡巨噬细胞(AM)上的表达,并探讨其作用机制及意义。方法:采用动物实验,以腹腔注射大剂量LPS诱导AL I模型;以腹腔注射生理盐水作为对照。用荧光定量逆转录—聚合酶链反应(RT-PCR)对AM表达的TREM 2 mRNA做定量分析。用EL ISA对支气管肺泡灌洗液中的肺肿瘤坏死因子(TNF-α)定量检测。结果:AM在正常情况下表达大量的TREM 2,在LPS造成急性损伤后表达大幅度下降。TREM 2与TNF-α的表达呈负相关(P<0.001)。结论:TREM 2可能在LPS致AL I中起重要的抗炎作用。
Objective:To investigate the expression of TREM2 in the alveolar macrophages(AM) of acute lung injury(ALl) induced by lipopolysaccharide (LPS) in mouse and to explore its mechanism. Methods ;By animal experiments ,the ALl models in mouse were induced by intraperitoneal injection of high-dose LPS. Controls were injected by intraperitoneal injection of normal saline TREM2 mRNA in lung tissues of mouse were quantitative analysis by real- time reverse transcription polymer-rase chain reaction(RT-PCR). ELISA was used to detect the concentration of TNF-α. Results:AM express a large number of TREM2 under normal circumstances ,but sharply descend after LPS caused acute injury. TREM2 expression was negative correlation with TNF-α expression (P〈 0. 001). Conclusion: TREM2 may play an important anti-inflammatory role in ALl caused by LPS.
