摘要
目的探讨扶正解毒颗粒(FJK)对镍诱导大鼠睾丸生精细胞凋亡的拮抗作用。方法选择健康成年Wistar雄性大鼠,NiSO42.5 mg/kg腹腔注射染毒,FJK 2.5、5.0和10.0 g/kg灌胃处理。30 d后,处死大鼠摘取睾丸,采用原位缺口末端标记法(TUNEL)检测生精细胞凋亡,免疫组化检测Bcl-2、Bax蛋白的表达水平。结果NiSO4染毒后,生精细胞总凋亡率升高(P〈0.05),以生精周期Ⅰ-Ⅺ期、Ⅻ-ⅩⅢ期和ⅩⅣ期的精原细胞和精母细胞为主(P〈0.05);精原细胞Bcl-2蛋白表达阳性细胞数减少,主要见于Ⅰ-Ⅵ期、Ⅻ-ⅩⅢ期和ⅩⅣ期(P〈0.05),精母细胞为Ⅰ-Ⅵ期、Ⅻ-ⅩⅢ期多见(P〈0.05);Bax蛋白表达阳性细胞数增多,精原、精母细胞均以Ⅰ-Ⅵ期、Ⅻ-ⅩⅢ期为主(P〈0.05)。FJK处理后,上述异常变化有所改善,以10.0和5.0 g/kg为主(P〈0.05)。结论FJK对NiSO4所致大鼠睾丸生精细胞凋亡具有拮抗作用,其机制可能与其引起Bcl-2表达上调和Bax表达下调有关。
Objective To explore the antagonistic effects of FuZhengJieDu granules(FJK) on the apoptosis of testis spermatogenic cells induced by nickel sulfate(NiSO4).Methods The male maturation Wistar rats were divided into five groups,including the control group with normal saline solution,NiSO4 2.5 mg/kg model group administrated intraperitoneally for 30 d,and FJK treated groups at doses of 2.5,5.0 and 10.0 g/kg with peroral route respectively for 21 days after the rats were treated with NiSO_4 2.5 mg/kg for 9 days.The apoptosis of spermatogenic cells and the expressions of Bcl-2、Bax protein were detected with TUNEL and Immunohistochemistry techniques.Results The number of apoptotic cells in NiSO4 model group was significantly higher than that of control group,and the apoptotic cells were mostly spermagonia and spermatocytes at the stages of Ⅰ~Ⅵ,Ⅻ~ⅩⅢ and ⅩⅥ during the spermatogenic cycle(P〈0.05).Compared with the control group,in spermagonia and spermatocyte of the seminiferous epithelium,the Bcl-2 protein expression was significantly decreased while Bax protein expression was obviously increased in NiSO_4 model group(P〈0.05).After treatment with FJK,the apoptosis rates of spermatogonia and spermatocyte were lower than those of the NiSO_4 model group in FJK 5.0,10.0 g/kg groups,and the Bcl-2 protein expression was significantly upward while Bax expression was downward in FJK 5.0,10.0 g/kg groups in the spermagonia and spermatocyte of the seminiferous epithelium(P〈0.05).Conclusion The results showed that FJK could antagonized distinctly the abnormal changes of spermatogenic cells apoptosis that could be related to the downward Bax expression and the upward Bcl-2 expression.
