摘要
目的探讨弓形虫感染对大鼠大脑组织神经丝(NF)mRNA表达和细胞免疫水平的影响。方法将4周龄雄性SD大鼠分成两组(每组10只):弓形虫感染组腹腔感染2×10^10个/L弓形虫速殖子悬液2ml:对照组腹腔注射灭菌生理盐水2ml。9周后,应用RT-PCR法检测大鼠大脑组织中高相对分子质量NF(NF-H)、中相对分子质量NF(NF—M)和低相对分子质量NF(NF—L)mRNA表达水平;流式细胞术检测大鼠外周血CD3^+、CD4^+、CD8^+T淋巴细胞;酶联免疫吸附试验测定其血清γ干扰素(IFN-γ)、肿瘤坏死因子α(TNF-α)、白细胞介素4(IL-4)水平。结果大鼠弓形虫感染9周后,大脑组织NF—L、NF—HmRNA表达量(0.51±0.06、0.68±0.05)较对照组(0.79±0.03、0.76±0.05)明显下降(t值分别为3.41、2.17,P〈0.01或〈0.05):NF—MmRNA表达量(0.69±0.02)与对照组(0.72±0.03)比较,差异无统计学意义(t=2.09,P〉0.05)。弓形虫感染组大鼠的CD4^+T细胞[(32.19±5.01)%]和CD8^+T细胞[(20.06±2.28)%1与对照组[(35.82±3.71)%、(22.06±3.90)%]比较,差异均无统计学意义(t值分别为1.69、1.88,P均〉0.05)。弓形虫感染组大鼠血清IFN-γ[(6.03±0.16)ng/L]、TNF-α[(18.05±1.93)ng/L]、IL-4[(15.76±1.59)ng/L]水平均高于对照组[(4.77±0.13)、(9.54±1.57)、(5.67±0.42)ng/L,t值分别为2.81、2.74、2.63,P均〈0.05]。结论弓形虫感染可导致大鼠大脑组织中NF亚单位mRNA表达水平下降。血清IFN-γ、TNF—α、IL-4水平升高。
Objective To investigate the alterations in levels of neurofilament (NF) mRNA and cellular immunity in rats infected with Toxoplasma gondii. Methods Four-week old male SD rats were randomly divided into two groups with ten animals for each. The subjects in the experimental group were i.p. injected with 2×10^10/L toxoplasma taehyzoits 2 ml for each. Meanwhile, each rat in the control group was i.p. injected with NaCl2 ml. After 9 weeks, mRNA levels of light molecular NF(NF-L), medium molecular NF(NF-M) and high molecular NF (NF-H) in cerebrum were determined by RT-PCR. The percentages of CD3^+、CD4^+、CD8^+ T lymphocytes were examined by means of flow cytometry, and the peripheral blood serum levels of IFN-γ, TNF-α, IL-4 were analyzed by ELISA. Results The mRNA levels of NF-L, NF-H in rat cerebrum(0.51 ± 0.06,0.68 ± 0.05) decreased than control group(0.79 ± 0.03,0.76 ± 0.05, t = 3.41,2.17, P〈 0.01 or 〈 0.05), the NF-M mRNA(0.69 ± 0.02) had no statistical significance(t = 2.09, P 〉 0.05 ) than control group(0.72 ± 0.03). No significant changes were observed on the levels of CD4^+T lymphocytes [ (32.19 ± 5.01 )% ] and CD8^+ T lymphocytes [ (20.06 ± 2.28 )% ], compare with the control group[ (35.82 ± 3.71 )%, (22.06 ± 3.90)%, t = 1.69,1.88, all P 〉 0.05]. The serum levels of IFN-γ[(6.03 ± 0.16)ng/L], TNF-α[(18.05 ± 1.93)ng/L], IL-4[(15.76 ± 1.59)ng/L] in experimental rats were significantly higher than those in the controls[ (4.77 ± 0.13), (9.54 ± 1.57), (5.67 ± 0.42)ng/L, t = 2.81, 2.74,2.63, all P 〈 0.05]. Conclusion Toxoplasma gondii infection can decrease NF subunit mRNA expression levels in rat brain tissue and increase serum levels of IFN-γ,TNF-α and IL-4 .
