摘要
目的制备特异性抗旋毛虫的鸡卵黄免疫球蛋白,研究其免疫应答规律及其稳定性。方法用纯化的旋毛虫幼虫免疫育龄种鸡,经水稀释法初步纯化浓缩后获得卵黄抗体。应用酶联免疫吸附试验检测卵黄抗体的效价及其敏感性和稳定性。结果通过免疫可成功诱导出高效价的抗旋毛虫抗体,效价可达1∶106以上,免疫应答持续时间可达34周以上。免疫后卵黄抗体敏感性和血清抗体敏感性无显著性差异(P>0.05)。稳定性试验表明IgY敏感性高,在pH4~10及温度不超过60℃条件下活性稳定。结论成功制备出高效价的特异性抗旋毛虫卵黄抗体,其敏感性高,有一定的耐热、耐酸碱性,可进一步用于旋毛虫感染的研究。
The specific chicken egg yolk immunoglobulin(IgY) was prepared and its activities was investigated in the present study,in which the purified antigen in Trichinella spiralis larvae was used to immunize hens together with Freund s adjuvant,the eggs laid from the immunized hens were collected and IgY was obtained was purified by water extraction,salt precipitation with ammonium sulfate and dialysis.The titer,sensitivity and stability of the chicken egg yolk antibody were determined by enzyme-linked immunoABSorbent assay(ELISA).It was found that the titer of antibody against T.spiralis produced by this method of immunization was above 1∶106 and the specific antibody activity lasted for more than 34 weeks.The sensitivity of IgY was just the same as that of antisera in serum(P0.05).IgY was stable under condition of pH4-10 and temperature above 60℃.It is evident that IgY produced after immunization in hens is proved to be highly tittered,sensitive and stable and it can be used for the further studies in the trichinella infection.
