安宫牛黄丸对脓毒症大鼠重要器官损伤及死亡率的影响 被引量:9

摘要 【目的】观察安宫牛黄丸对脓毒症大鼠重要器官损伤及死亡率的影响。【方法】选用SD大鼠,随机分为正常对照组,模型组,安宫牛黄丸低、中、高剂量组(中药低、中、高剂量组,剂量分别为1.0、2.0、3.0 g.kg-1.d-1)。除正常对照组外,其他组均采用盲肠结扎穿孔(CLP)法复制腹腔感染型脓毒症模型,中药低、中、高剂量组分别于造模前0.5 h和造模后6 h灌胃给药,观察24 h大鼠死亡率,检测造模24 h后各组大鼠血清丙氨酸氨基转移酶(ALT)、总胆红素(TB)和肌酐(Cr)含量及肺组织髓过氧化物酶(MPO)活性。【结果】模型组血清ALT、TB水平,肺组织MPO活性及24 h死亡率均显著升高(P<0.05或P<0.01)。中药低、中、高剂量组可显著降低肺组织MPO活性(P<0.05或P<0.01),其中低剂量组可显著降低血清TB水平(P<0.05),低、中剂量组还可显著降低24 h死亡率(P<0.05)。【结论】安宫牛黄丸可降低脓毒症大鼠的死亡率,并对肺、肝等重要器官有一定的保护作用。 Objective To observe the effects of Angong Niuhuang Bolus (ANB) on vital organ injury and mortality in rats with sepsis. Methods SD rats were randomized into normal control group, model group, and low-, middle- and high-dose ANB groups ( in the dose of 1.0, 2.0 and 3.0 g . kg^-1.d^-1 respectively). Except that the normal control group, the rats in other groups received cecal ligation and puncture (CLP) method to induce sepsis through abdominal infection. ANB groups were given gastric gavage of ANB 0. 5 hours before CLP and 6 hours after CLP. After CLP for 24 hours, the mortality was observed, serum levels of alanine aminotransferase (ALT) , total bilirubin (TB) and creatinine were detected, and pulmonary, myeloperoxidase (MPO) activity in all of the rats was examined. Results Serum ALT and TB levels, MAO activity and 24-hour mortality were increased in the model group (P 〈0. 05 or P 〈0. 01 compared with those in the normal control group). Low-, middle- and high-dose ANB decreased MAO activity (P 〈0. 05 or P 〈0. 01 compared with that in the model group), low-dose ANB decreased serum TB level (P 〈 0. 05 ), and low- and middle-dose ANB decreased 24-hour mortality obviously in sepsis rats ( P 〈 0.05). Conclusion ANB can decrease the mortality of rats with sepsis, and has certain protective effect on vital organs such as lung and liver.
