摘要
用高效液相色谱法测定血清色氨酸。采用Nova-PAKC18柱,0.1mol·L-1磷酸二氢钾水溶液为流动相,295nm的检测波长,测出线性范围为1.223~2447.381μmol·-1,最低检出浓度为0.612μmol·L-1,正常人血清色氨酸浓度为46.603±5.108μmol·L-1。测定血清色氨酸的时间<5min。本法简便、快速、准确、可靠。
In this paper a high performance liquid chromatographic(HPLC) method has been developed to determine the concentration of tryptophan(Trp) in human serum. Serum sample was precipitated with trichloroacetic acid solution(0.6mol·L -1 ) to remove protein and centrifuged and assayed by HPLC. The operating conditions were NOVA PAK C18 column(3.9mm×150mm), KH 2PO 4(0.1mol·L -1 ) mobile phase with flow rate of 0.7ml·min -1 , and detective wave length at 295nm. The linear range was 1.223~2447.381μmol·L -1 . The detection limit was 0.612μmol·L -1 . The average recovery was 96.3%. Intra assay CV is less than 4% and inter assay less than 5%. The concentration of Trp in normal adult serum was 46.603±5.108μmol·L -1 . The detection time of serum Trp was within 5min. The results suggest that this method is simple, fast, accurate, and convenient.
