Establishment and application of PCR-SSP for detecting Mur blood group

摘要 目的建立Mur血型的分子生物学诊断方法。方法根据Mur血型的分子背景,设计特异性引物,采用PCR-SSP技术,优化反应体系及扩增参数,建立Mur血型分子检测方法;同时采用标准抗血清、标准细胞,用血清学方法予以验证并初步应用于献血者Mur血型的检测。结果建立了Mur血型PCR-SSP检测方法,用以检测14份已知抗-Mur阳性或阴性的献血者标本,同时用血清学方法平行检测:抗-Mur阳性11/11、抗-Mur阴性3/3,结果完全一致;用本法对随机抽取的107名献血者血样检测,检出2例阳性,同时采用标准抗血清用血清学方法所做的复核结果也完全一致。结论所建立的Mur血型PCR-SSP检测方法可以准确地检出Mur抗原,弥补了Mur血型血清学检测方法的局限性,有助于Mur血型检测方法的标准化、规范化。 Objective To establish the molecular detection method of Mur blood group. Methods To design Mur gene sequence specific primers according to the molecular background of Mur blood group and establish the PCR-SSP to de- tect Mur blood group. The conditions of PCR reaction was optimized. This method was validated by standard samples and applied in Mur blood group detection of blood donors. Results The molecular method of Mur blood group was established. And this methods was validated by standard samples(P 〈 0.01 ). A total of 107 random donors were detected parallel Mur blood group by this method and serology test. The results of this method showed that there was no statistically significant difference with serology test(P 〈 0〈 01 ). Conclusion The molecular method of Mur blood group can be applied in detection of Mur blood group.
