摘要
目的探索一种用线栓法制作稳定的大鼠单侧大脑中动脉阻塞(MCAO)模型的简易方法。方法不分离结扎翼腭动脉(PPA),用3种不同的线栓即头端未处理的鱼线、指甲油处理的鱼线和特制的尼龙线栓,分别经大鼠的颈总动脉(CCA)和颈内动脉(ICA)插线,分析不同种类线栓和插线部位对大鼠单侧MCAO手术的插线时间及建立MCAO模型成功率的影响;观察2种不同插线部位和结扎颈外动脉(ECA)与否对大鼠MCAO建模成功率的影响。结果3种线栓的插线时间比较差异无统计学意义(P=0.397);未处理的鱼线的建模成功率低于指甲油处理的鱼线和特制的尼龙线栓(P=0.001),而后2种线栓的建模成功率比较差异无统计学意义(P=0.626);经CCA插线时间明显低于经ICA插线时间(P=0.002);两者的建模成功率比较,差异无统计学意义(P=0.441);2种不同部位插线的建模成功率比较及结扎ECA和未结扎ECA的建模成功率比较,差异均无统计学意义(均P=0.694)。结论采用指甲油处理头端的鱼线,结扎ECA经CCA插线可以简单快捷地建立稳定的大鼠单侧MCAO模型。
Objective To explore the simple approach to make the steady middle cerebral artery occlusion(MCAO) model in rats by using suture embolization method. Methods In this study,we didn't isolate and ligate the pterygopalatine artery,and used three kinds of suture-the untreated fishing thread,the fishing thread with its terminatio treated with varnish and the specially made nylon suture, inserting from the common carotid artery(CCA) and the internal carotid artery (ICA) respectively,to analyze the effect of different kinds of sutures and inserting positions to the time of insertion and the success rate of the model building;We observed the effect of the two different positions of inserting the suture from and ligating the external carotid artery(ECA) or not to the success rate of the model building. Results The time of insertion among the three kinds of sutures have no significant difference(P=0.397). The untreated fishing thread's success rate of the model building was lower than the fishing thread's with its terminatio treated with varnish and the specially made nylon suture's(P=0.001), and the rates between the latter two had no significant difference(P=0.626). The time of insertion from CCA is obviously shorter than that form ICA (P=0.002); And the success rates between the two had no significant difference (P=0.441);The success rates of the model building between the two different inserting positions had no significant difference(P=0.694)and also the rates between ligating the ECA and non-ligating the ECA(P=0.694). Conclusion Applying the fishing thread with its terminatio treated with varnish,ligating the ECA and inserting from the CCA could build steady unilateral MCAO model in rats simply and quickly.
