摘要
目的:探讨人脑胶质瘤中wnt1、β-Catenin、Ki-67的表达及与细胞增殖的相关性。方法利用组织芯片技术用及免疫组化方法检测wnt1、β-Catenin、Ki-67在正常脑组织、低/高级别星形细胞瘤中的表达,并分析其相关性。结果正常脑组织中wnt1、β-Catenin、Ki-67表达均为阴性或弱阳性。wnt1、β-Catenin、Ki-67的表达均随胶质瘤级别增高而增高,表达与组织分级密切相关(P<0.01),β-catenin在Ⅰ级胶质瘤中中阳性率33.3%(2/6),阳性部位为胞质;Ⅱ级阳性率28.6%(8/28),阳性部位为胞质;Ⅲ级阳性率60.0%(18/30),阳性部位为胞质,其中3例出现核阳性;Ⅳ级阳性率78.3%(18/23),其中8例出现核阳性。wnt1、β-Catenin、Ki-67表达的表达两两相关(P<0.01)。结论β-Catenin、Ki-67的异常表达与胶质瘤的增殖活性增高有关,其与wnt1相互协同促进胶质瘤的发生发展,应用组织芯片进行胶质瘤分子病理学研究较常规免疫组化方法与具有明显优势。
Objective To investigate the expression of wntl, β-Catenin, Ki-67 in gliomas and their relationships with cell proliferation in gliomas. Methods This study comprised 6 cases of normal brain tissues and 94 cases of glioma tissues. Tissue microarray and Immunohistochemistry were employed to detect the expression of wntl,β-Catenin, and Ki-67. Results The expressions of wntl,β-Catenin, and Ki-67 increased, which was associated with the degree of malignancy of tumors. The β-Catenin positive expression rate was 33.3% (2/6)in grade Ⅰ , 28.6% (8/28)in grade Ⅱ, 60.0 % ( 18/30) in grade Ⅲ, and 78.3 % (18/23) in grade Ⅳ respectively. The differences of positive rate and expression level between the low-grade and high-grade tumors were statistically significant. In addition, the expression of wntl, β-Catenin, and Ki-67 was correlated eath other(P 〈 0. 01). Conclusion Up-regulation of wnt1 , β-Catenin and Ki-67 expression can be used as an important histological sign for the diagnosis and identification of human tumor. β-Catenin,and Ki-67 over-expression would be related to gliomas proliferation. And tissue microarray provides a rapid and powerful histological method that may be used in immunohistochemistry and plays an important role in gliomas biology.
