摘要 目的筛选戊型肝炎病毒优势表位抗原以便研发新型戊型肝炎诊断试剂。方法通过计算机辅助软件分析,利用改构的高效表达载体表达了HEV 1型与HEV 4型ORF2和ORF3的17种表位抗原,利用蛋白芯片技术平台筛选优势表位抗原。结果在所获得的17种表位抗原中筛选到4种可以作为研发新型戊型肝炎诊断试剂候选抗原的优势表位抗原。结论利用蛋白芯片技术快速准确地筛选到4种可用于研发新型戊型肝炎诊断试剂的候选优势表位抗原。 Objective Screening epitope-dominant antigens of Hepatitis E Vires for prepare the New diagnosis Kit. Methods The epitopes of HEV I and iV ORF2, ORF3 were analyzed by the BIOSUN software. Seventeen epitope antigens were cloned, expressed and purified. The epitope-dominant antigens of Hepatitis E Virus were screened using the protein microarray. Results The four epitope-dominant antigens were picked out from the 17 epitope antigens, which were the candidate epitope-dominant antigens for developing the newly anti-HEV diagnosis reagent. Conclusion Through the rapid and accurate screening of epitope-dominant antigens of Hepatitis E Virus by the multi-target protein microarray,4 epitope-dominant antigens were picked out for developing the newly anti-HEV diagnosis reagent.
