摘要
三氧化二砷是治疗急性早幼粒细胞白血病的特效药。三价砷是巯基(-SH)的络合剂,砷化物与-SH结合后可能通过多条途径诱导细胞凋亡,此外,砷与-SH结合还直接导致细胞内的还原性谷胱甘肽耗竭,使活性氧产生增多。研究发现,还原性谷胱甘肽(GSH)水平与三氧化二砷对急性早幼粒细胞白血病、多发性骨髓瘤、淋巴瘤等细胞株的疗效呈负相关,降低细胞内GSH水平会明显增强三氧化二砷的疗效。
Arsenic trioxide is crucial in treating acute promyelocytie leukemia (APL). Arsenic trioxide, the complex compound of sulfhydryl (-SH), may induce apoptosis by many pathways after combination with-SH. In addition, this combination could increase the level of reactive oxygen species(ROS) by depleting intraeellular glutathione(GSH) directly. It is found that there is a negative correlation between the level of GSH and the effect of arsenic trioxide on cell lines treatment of APL,myeloma(MM) and lymphoma, that is to say, decreasing the level of intracellular GSH could tremendously enhance the treatment effect of arsenic trioxide.
