T7 Expression System and Protein Production by Auto-induction Strategy

摘要 目前较为通用的重组蛋白表达策略即为诱导T7表达系统,该系统能较好地控制并表达出大量的蛋白产物,但该系统仍存在一定的不足。近几年,Studier及其同事们提出了自诱导的方法,他们提供的自诱导表达策略无论在产出蛋白的质量还是数量上都比传统方法有很大提高。 A very popular method of ensuring tight control, as well as high-level induced expression, is to place the gene to be expressed in a plasmid under control of a T7 phage promoter. However, there s surely some disadvantages in the inducible expression systems which were currently used. Studier and his coworkers invented the concept of auto-induction, using their auto-inducing media in high-density shaking cultures might get proteins better both in quality and quantity.
