摘要
用十二烷基磺酸钠法分别提取玄参新鲜根及干燥根的DNA.电泳结果显示,从玄参新鲜根中可提取到完整DNA,而从"发汗"后的玄参干燥根中提取的DNA已降解.使用紫外分光光度法对新鲜根中提取的DNA进行纯度鉴定,A260/A280为1.83.结果表明,所采用的十二烷基磺酸钠法可经济高效地从玄参根部提取到高质量的DNA.
Scrophularia ningpoensis Hemsl.is a typical tradtitonal Chinese herb,which contains phenolics,terpenes and glycoside,et al.In this paper,the modified methods of SDS were used to extract genomic DNA from fresh roots and desiccated roots respectively.The results show that the genomic DNA can extract from fresh roots,but can not extract from desiccated roots.And the A260/A280 of genomic DNA from fresh roots is 1.83.In conclusion,this provides an economic,convenient and effective method of DNA isolation for a biological study of scrophularia ningpoensis Hemsl.
