摘要
[目的]探讨提取海南绿茶籽茶皂素的最佳工艺条件,分析茶籽油成分。[方法]以海南绿茶籽为原料,采用加热回流法和微波辅助提取绿茶籽中的茶皂素,并将其提取结果进行比较。采用正交试验筛选最佳提取工艺条件,并用GC-MS对茶籽油成分进行分析。[结果]正交试验表明,影响茶皂素提取率的因素依次为:温度>无水乙醇>辐射时间>微波功率;微波法提取海南绿茶籽茶皂素的最佳提取工艺条件为:绿茶籽质量与无水乙醇体积比为40∶300(g/ml)微波提取功率为600W,微波提取温度为50℃,提取时间为50min,在该条件下,茶皂素得率为47.89%。海南绿茶籽油经GC-MS分析鉴定出8种化合物,以酸类化合物为主。[结论]与加热回流提取法相比,采用微波法缩短了提取茶皂素的时间,提高了茶皂素的得率。
[ Objective ] The study aimed to explore the optimum process conditions of extracting tea saponin from the green tea seed and analyze the components of tea seed oil. [ Method ] With Hainan green tea seed as raw material, the tea saponin was extracted from the Hainan green tea seed by using heating reflux method and microwave-assisted extraction, and their extraction results were compared. The optimum process conditions of extracting were screened out by using orthogonal test ,and the components of tea seed oil were analyzed by GC-MS. [ Resuit] The orthogonal test experiment showed that the factors of affecting the extraction rate of tea saponin in order were as follows:extraction temperature 〉 anhydrous alcohol 〉 irradiation time 〉 microwave power. The optimal process conditions of extracting tea saponin from Hainan green tea seed were:the ratio of quality of green tea to volume of anhydrous alcohol was 40:300( g/ml), microwave power was 600 W,microwave extraction temperature was 50 ℃, extraction time was 50 min, under these technological conditions, the yield of tea saponin was 47.89%. Eight compounds were identified from the tea seed oil by GC-MS, mainly with the acids compounds. [ Conclusion] Compared with the heating reflux method ,the microwave-assisted extraction method could shorten the extraction time of tea saponin and increase the yield.
