靶向人端粒酶小干扰RNA对人喉癌Hep-2细胞系形态学影响

Effect of siRNA by targeting the human hTERT gene on morphology in Hep-2 cells

摘要 目的探讨靶向人端粒酶基因siRNA对人喉鳞状细胞癌Hep-2细胞形态学的影响。方法根据人端粒酶基因设计和合成人端粒酶siRNA和随机的siN.C作为阴性对照,通过转染载体Lipofectamine 2000脂质体转染Hep-2细胞,观察形态学结构。结果转染siRNA48h后细胞皱缩、变圆并开始脱落。HE染色显示细胞呈明显的良性转化。透射电镜下可见细胞凋亡、胀亡的形态改变。结论靶向人端粒酶基因的siRNA能促使Hep-2细胞形态呈良性转化,并能促进肿瘤细胞凋亡。 Objective To investigate the effect of the small interfering RNA (siRNA) targeting hTERT gene in Hep-2 cells on morphology. Methods siRNA targeting site 1718 of hTERT mRNA was designed and synthesized. In vitro cultured Hep-2 cells were transfected with lipofectamine 2000 and the inhibitory effect was observed by phase contrast microscopy and transmission electron microscopy. Results 48 hours after siRNA were transfected into Hep-2 cells,numerous round cells and shrinkage of the cellular membrane were observed under phase contrast microscopy, cells showed innocence transformation under HE stain microscopy, and apoptosis and oncosis appeared under transmission electron microscopy. Conclusion siRNA can promote the Hep-2 cell to show innocence transformation and apoptosis.
